The week ended. It is the very end of my busy week. My final year project’s report is handed in, that’s really good news. And there is also the web application assignment which has also ended (although I didn’t complete the web application that I was required to in the end.
This week, I haven’t been sleeping well. Everydy, I sleep at around 2 a.m. and then wake up at 7 a.m.. It’s usuaully 5 hours of sleep me. All things have ended. I can’t say it ended off very well though.
Today, I went to take a test paper on genomic techniques without even studying for it even a bit. I haven’t even seen the notes the past week.
So without any idea what the entire topic is about, I went for the test. When I reach the classroom, I ask Hai Jiao what does YAC stand for. That’s the only thing I have in mind during that time. I only know that YAC stands for “Yeast Artificial Chromomsome” and nothing else. That’s how bad it was. I did listen to class a little here and there, but wspend the other times talking or probably even missing classes.
The tested started. I looked at the paper and got a shock. What’s wrong? Everything. Nearly. I don’t know anything. Everything seemed so unfamiliar. And the first question read something like, “Give 3 reasons why YAC is used in place of eukaryotic chromsome.”
Huh?
Yeah, that’s what I thought. I only know what it stands for! So I wrote something like this:
YAC stands for Yeast Artificial Chromosome.
YAC can contain DNA insert (this is bullshit, it’s like everyone knows that).
YAC has origin of replication.
YAC has many, many sites that replication can occur.
That’s total crap! Then there was this question on what is nick translations.
Nick translation is when the mRNA transcripts are translated into protein sequences instaneously as they are transcribed from the DNA sequence. Such coupled process is known as “nick translations”
BULL SHIT! It’s wrong, totally wrong. Now, would some smart guy tell me what this “nick translation” is all about? Don’t tell me about English-French translation of “Mr. Dew”.
And when I’m doing the test paper, for once, I felt relaxed. Not because I am confident, but because I can crap all I want for the day. I got amused and smiled while scribbling too! I’ll update you on what’s my results for that test, lol.

Ok, nick translation definition:
“A procedure for making a DNA probe in which a DNA fragment is treated with DNase to produce single-stranded nicks, followed by incorporation of radioactive nucleotides from the nicked sites by DNA polymerase I.”
DNA labeling can also be done by nick translation.
Anyways that is all I know. Ask me no further than this.
On an off topic note, I’m sorry the comment you left in my site suddenly disappeared. The ISP that is hosting my site screwed up the database and probably reinstated a backup that was done before 4AM. It affected another sub-domain I was hosting. But anyways thank you for the comment.
